چکیده
Background Salivary glands from blood-feeding arthropods secrete several molecules that inhibit mammalian hemostasis and facilitate blood feeding pathogen transmission. The salivary functions Simulium guianense , the main vector of Onchocerciasis in South America, remain largely understudied. Here, we have characterized a protease inhibitor (Guianensin) blackfly . Materials methods A combination bioinformatic biophysical analyses, recombinant protein production, vitro vivo experiments were utilized to characterize molecula mechanism action Guianensin. Kinetics Guianensin interaction with proteases involved vertebrate inflammation coagulation carried out by surface plasmon resonance isothermal titration calorimetry. Plasma recalcification coagulometry tail bleeding assays performed understand role coagulation. Results was identified sialotranscriptome adult S. flies belongs Kunitz domain inhibitors. It targets various serine inflammation. Binding these enzymes is highly specific catalytic site not detectable for their zymogens, site-blocked human factor Xa (FXa), or thrombin. Accordingly, significantly increased both PT (Prothrombin time) aPTT (Activated partial thromboplastin plasma consequently clotting time ex also inhibited prothrombinase activity on endothelial cells. We show acts as potent anti-inflammatory molecule FXa-induced paw edema formation mice. Conclusion information generated this work highlights biological functionality an antithrombotic may play significant roles

متن کاملImmunosuppressive, anti-inflammatory and anti-cancer properties of triptolide: A mini review
Objective: Triptolide, the active component of Tripterygium wilfordii Hook F has been used to treat autoimmune and inflammatory conditions for over two hundred years in traditional Chinese medicine. However, the processes through which triptolide exerts immunosuppression and anti-inflammation are not understood well. In this review, we discuss the autoimmune disorders and inflammatory condition...

Objectives: Aim of the present work was to investigate the anti-inflammatory, analgesic and antiulcer effects of red seaweed Porphyra vietnamensis (P. vietnamenis). Materials and Methods: Aqueous (POR) and alcoholic (PE) fractions were successfully isolated from P. vietnamenis. Further biological investigations were performed using a classic test of paw edema induced by carrageenan, writhing in...
متن کاملAnti-inflammatory and anti-oxidant properties: Is there a link?
BACKGROUND It is believed that the anti-inflammatory activity of medicines is closely related to their antioxidant activity. However, in clinical practice rigorous evidence-based medicine approach fails to reveal important effects of antioxidants on patient important outcomes in inflammatory disorders, as has been shown by a number of Cochrane reviews [1-3]. OBJECTIVE To evaluate anti-inflamm...
متن کاملMXRA5 is a TGF‐β1‐regulated human protein with anti‐inflammatory and anti‐fibrotic properties
Current therapy for chronic kidney disease (CKD) is unsatisfactory because of an insufficient understanding of its pathogenesis. Matrix remodelling-associated protein 5 (MXRA5, adlican) is a human protein of unknown function with high kidney tissue expression, not present in rodents. Given the increased expression of MXRA5 in injured tissues, including the kidneys, we have suggested that MXRA5 ...
